WebFeb 28, 2024 · Hofmann Rule. Hofmann rule is an empirical rule used to predict regioselectivity of 1,2-elimination reactions occurring via E1cB mechanism. It states that, in a regioselective E1cB reaction, the major product is the less stable alkene, i.e., the alkene with the less highly substituted double bond. For example see Hofmann elimination. WebZaitsev’s consolidation of data on the rates of base elimination reactions led to his formulation of Zaitsev’s rule predicting the regiochemistry of the reaction. Hofmann’s studies of the pyrolysis of quaternary ammonium ions revealed a complementary regiochemical outcome, which he formulated in the Hofmann Rule.
